چکیده

Vitamin K is a substrate for the enzyme catalyzing the carboxylation of specific glutamyl residues to y-carboxyglutamy1 residues in hepatic precursors of a limited number of plasma proteins, including prothrombin. The y-carboxylation of these proteins can be blocked by the anticoagulant warfarin; and in the bovine and human, warfarin treatment results in the secretion of under-y-carboxylated forms of prothrombin into plasma. In the rat, this response is not seen, but plasma prothrombin concentrations are drastically decreased. This response has now been studied in rat hepatoma (H-35) cells in which prothrombin secretion is decreased 90% by incubation in the presence of warfarin. Neither prothrombin mRNA levels nor the apparent rate of prothrombin message translation were decreased when cells were cultured in the presence of warfarin rather than

Prothrombin synthesis and degradation in rat hepatoma (H-35) cells: effects of warfarin.

Vitamin K is a substrate for the enzyme catalyzing the carboxylation of specific glutamyl residues to gamma-carboxyglutamyl residues in hepatic precursors of a limited number of plasma proteins, including prothrombin. The gamma-carboxylation of these proteins can be blocked by the anticoagulant warfarin; and in the bovine and human, warfarin treatment results in the secretion of under-gamma-car...

HEPATOMA McARDLE-RH7777 CELLS HAVE THE SAME RESPONSE AS NORMAL RAT HEPATOCYTES TO BOTH DIBUTYRYL-cAMP AND ANTICALMODULINW-7

The effects of cAMP-analogue dibutyryl-cAMP and anticalmodulin W-7 were studied on de novo synthesis and secretion of lipids in cultures of hepatoma McArdle RH7777 cells and normal rat hepatocytes. Dibutyryl-cAMP and W -7 separately caused a significant decrease in the secretion of de novo synthesized triacyl [3H]glycerol in both cultures of McArdle cells and rat hepatocytes. The inhibito...

Induction of prothrombin synthesis by prothrombin fragments.

The mechanisms by which blood levels of prothrombin (PT) are regulated in the vitamin K-sufficient state are unknown. We have studied PT synthesis by Reuber H-35 rat hepatoma cells exposed to vitamin K and [3H]leucine in serum-free cultures. Administration to the culture system of exogenous bovine PT and rat PT was characterized by increases in endogenous PT synthesis and secretion of 2- and 3-...

Effect of warfarin on prothrombin synthesis and secretion in human Hep G2 cells.

Prothrombin synthesis and secretion were studied in a human hepatoma cell line (Hep G2) incubated with 35S-methionine for 2 to 24 hours at 37 degrees C. Extracellular and intracellular prothrombin were detected by immunoprecipitation with affinity-purified antiprothrombin antibody. Incorporation of 35S-methionine into prothrombin was monitored by counting specific bands excised from 10% sodium ...

Structural features of the kringle domain determine the intracellular degradation of under-gamma-carboxylated prothrombin: studies of chimeric rat/human prothrombin.

Vitamin K antagonists such as warfarin inhibit the vitamin K-dependent gamma-glutamyl carboxylation during protein processing and block the secretion of under-gamma-carboxylated prothrombin (FII) in the rat but not in the human or bovine. Under-gamma-carboxylated prothrombin is also secreted from warfarin-treated human (HepG2) cell cultures but is degraded in the endoplasmic reticulum in warfar...
